Pharmacokinetic and Pharmacodynamic (PK/PD)

Resource Director: Robert DiPaola, MD

PK and PD studies are critical to cutting edge phase I and pilot trials involving evaluation of new therapeutic agents, and in the development of new drug combinations. In particular, PK/PD studies are used to define optimal doses and schedules of administration for new compounds with limited toxicity.

The PK/PD Shared Resource of CINJ provides a centralized, high quality, cost-effective facility for performing clinical and preclinical pharmacologic studies.

Location
The PK/PD Shared Resource is located in CINJ Lab 2050.

Objectives
The specific objectives of this shared resource are to:

  • to assist investigators in clinical pharmacologic studies of cancer therapeutics
  • to assist investigators in preclinical studies of cancer therapeutics

Services

  • designing blood, urine, and/or tissue sampling strategies based upon available preclinical and clinical pharmacology
  • developing GLP-based analytical methods for molecules of interest
  • quantification of compounds in blood, urine, saliva, and tissue from animals or human, employing HPLC or mass spectrophotometric methods
  • PK modeling of concentration-time data
  • determining relationships between PK parameters and PD variables, including clinical and tissue specimen data. Assistance is given to investigators in designing sampling strategies for assessment of drug targets or potential biomarkers in normal and malignant tissues
  • preclinical murine toxicity and pharmacokinetic studies

Correlative statistical analyses are performed in collaboration with the Biometrics Shared Resource.
