The steadily expanding use of genetically engineered mouse models in cancer research has resulted in an explosion of information on the in vivo functions of oncogenes and tumor suppressor genes, with important consequences for pre-clinical testing and potential therapeutics. To support research with genetically engineered mice at CINJ, the Transgenic/Knock-out Mouse Shared Resource provides a centralized, expert facility for the generation of genetically engineered animals and assists CINJ investigators with their design and analysis.   

The specific objectives of this shared resource are:

  1. production of transgenic mice through pronuclear microinjection of DNA constructs;
  2. production of gene-targeted mice through comprehensive services for ES cell culture, transfection, and screening, as well as generation of chimeric mice by blastocyst injection of targeted ES cell clones. Expert consultation with investigators will be provided for targeting strategies and overall experimental plan;
  3. molecular biology services associated with the production and handling of genetically engineered mice, including preparation of DNA samples for pronuclear microinjection, assistance with screening of ES clones for targeted events, and genotyping of mice;
  4. assistance with mouse husbandry and phenotypic analysis through education and training for inexperienced investigators in the fundamentals of mouse handling and breeding strategies, as well as consultation on methods for phenotypic analysis of mouse mutants;
  5. storage of mouse strains through sperm and embryo cryopreservation; and
  6. re-derivation of mouse strains for importation into specific pathogen-free (SPF) barrier housing.

In addition, in the future this shared resource will not only provide a service function but will also play an important intellectual role in the development and pursuit of mouse-oriented research projects in the CINJ community.
